Effect of pyridoxal phosphate on atherosclerosis and nephropathy progression in the atherosclerotic rats

Background and Objectives: Diabetes vascular complications are the principal cause of death in the world. Therefore, we investigated the effect of pyridoxal phosphate (PLP) on the formation of atheromatous plaque formation and renal function in the atherosclerotic rats. Materials and Methods: Male Wistar rats were randomly divided into 4 equal groups (ten rats in each group): normal, atherosclerotic, and two alike groups under PLP treatment. Atherosclerosis induced in rats with an atherogenic diet. Treated groups of rats, normal and atherosclerotic, were treated with daily 0.18 % of PLP in drinking water for 3 months. Hematoxylin–Eosin (H&E) stains were employed to survey the histopathological changes of the aorta. Insulin resistance index, the activity of glyoxalase system, lipid profile, LDL oxidation markers, advanced oxidation protein products, and inflammatory markers (hs CRP and TGF-β1), as well as creatinine in the serum, and protein excretion in the urine of all rats was determined. Results: Atheromatous lesions were not observed in the aorta of the PLP treated atherosclerotic rats. Furthermore, PLP showed an advantageous effect on insulin function, lipid profile, kidney function parameters, glyoxalase system activity, and inflammation. Over and above, the treatment decreased the formation of LDL oxidation products, both in vitro and in vivo (p > 0.001). Conclusion: PLP showed a beneficial effect on vascular complications in atherosclerotic rats owing to antioxidant, and anti-inflammatory properties in addition to the beneficial effects on insulin function, dyslipidemia, and glyoxalase system activity
